BODIPY vs. FITC

We have compared these fluorochromes (BODIPY and FITC) when tagged to
dUTP, as markers of DNA strand breaks (Cytometry, 20:172, 1995).
BODIPY was somewhat better in terms of fluorescence intensity (30%
higher fluorescence) using the standart filter setup in FASCan. The
major diffrence, however, was much lower rate of BODIPY fluorescence
fading when we viewed the cells under UV light microscope (blue
filter) and attempted to make photographs. There was hardly any loss
of BODIPY fluorescence during e.g. 20 sec exposure wheras FITC
fluorescence was visibly decreased.
Another advantage of BODIPY is the narrower band of fluorescence
emission (compared to FITC), which is helpful in multicolor staining
combinations.
