Re:PI and 3 colour mouse lymphs

We have used anti CD8a-Red613 or anti-B220-Tricolour with PI. By
titrating the amount of PI used, you can separate the population from
the antibody stained ones. Usually PI staining is 5-10x brighter. In the
first case the PI+ will show up in the (red613-)corner of a PE/red613
plot, but will appear as a double positive population if you use TC as a
third colour, since less compensation is needed. We usually stain lymphs
and set amplifiers to find them around 7x10e2 on our FACSort-Cellquest
set up. PI positives are usually found above 3x10e3.
