Kirk,
There is a single-step method for AO determination of DNA/RNA in Taylor, I.W. and Milthorpe, B.K.
An evaluation of DNA fluorochromes, staining techniques and analysis for flow cytometry: 1. unperturbed cell populations.
J. Histochem. Cytochem. 28; 1224, 1980.

It works well on lymphocytes, but the conditions are fairly critical, as with
all AO methods.
Give it a go and email me if it doesn't work!
