I would like to stain T cells from a Mixed Leukocyte Culture(MLC) using
three
different antibodies. The antibodies I have used in the preliminary
experiments is FITC-CD8, anti-Vbeta-biotin+red 670 avidin and PE-CD44.
However, there seem to be problem with the CD44 antibody, it stains only
were few cells. Is it a bad antibody to chose if you want to study the
activated T cells from MLC in particular? Any alternative antibodies? If I
stain Con A activated T cells with the CD44 ab. it give a clear positive
population. I have also done titration of the antibody in order make sure
that a use the right amount of antibody.
I would appreciate if someone could give me some advice.
Yours sincerly,
