Indo-1 in CHO cells

Greetings,

One of the labs here is trying to measure calcium
flux in CHO and CHO transfectants with INDO-1 but
the results have been somewhat less than spectacular.
The amount of dye is not unreasonable (compared to
Jurkats) although lower but the signal we are getting
with ionophore is drastically reduced. My suspicion
would point to a lowered esterase activity but befoe
I go to any great lengths I wanted to check to see if
anyone might have run across this problem in these
cells before and be able to offer some suggestions.
