Dear flow-ers,
I had asked some advices on performing cytoplasmic IgM staining and the
sources of antibodies for both human IgM and malaria-infected. I have
gotten several good advises on these questions and would like to summerize
the info that I gathered.
1. Antibody for human IgM: The following antibodies all work well. Goat
anti human IgM from Caltag, Mouse anti human IgM from BD, anti-human IgM
from Dako and anti-human IgM from Tago.
2 Regarding fixation and permeabilization of cell for cytoplasmic
staining, researcher have been using either Fix and Perm from Caltag
(suggestion from Dr. Bob Johnson), FACS permeabilization solution from BD,
a published formula of three solutions from Pharmingen homepage, or An der
Grub Lysing reagent (from Dr. C. Thomas Nebe). We have try using both the
reagents from Caltage and BD. They worked equally well in our hands.
3. regarding antibody for malaria, We got a very good advice from Alan
Hibbs from Queensland Institute of Medical Research, P.O. Royal Brisbanw
Hospital. From his advise, we learned that it is not feasible to screen
blood donors for malaria infection regardless of the method one may use.
Also, there is no commercial source for anti-malaria antibodies. Thank you,
