I have tried Hoechst as well as DAPI with thiazole orange after fixing with
1% glutaraldehyde plus 0.03% SDS. Thiazole orange discriminated well between
RBCS, reticulocytes and nucleated cells, but Hoechst or DAPI was not picked
up well enough to identify micronuclei by microscope or flow cytometer,
although, It did stain nucleated cells. When I tried Acridine orange (AO),
micronuclei were intensely stained (microscopic observation), but I want to
stay away from running AO through my machine.
I appreciate any help on this subject.
