We routinely use Hoechst in our laboratory. We published a
fairly thorough investigation of the dye properties, Scatchard and
Swartz plots comparing various fixations, concentrations etc. of
Hoechst 33258 to 33342 (J. Histochem.Cytochem. 36:757-762).
We now also know that the cells can also fixed in 0.5-2%
paraformaldehyde (PF) for 15 minutes followed by 25%buffered
ethanol with no effect on the binding curves (we only
investigated Hoechst 33258 for this). Higher concentrations of
PF introduce higher CV's and begin to distort other features of
interest. Our current protocol for Hoechst dna in multiple
marker assays can be found in this month's PNAS 1993, 8287-
8290.
Please feel free to contact me if I can be of any further
assistance.
