Hi, Peter,

I haven't actually used COS cells, but we had problems loading CHO cells,
which pump indo-1 out. You should definitely try using the Pluronic, though
it might not help much. Verapamil will probably help a lot, but it is a
blocker of voltage-gated calcium channels. That probably won't matter for
your cells, but you should be aware of that. We usually load CHO cells while
they are attached, but they can also be loaded in suspension. We ended up
isolating a mutant CHO cell line that loads well with indo to do our studies
(see report in Cytometry 17:33-38, 1994). Hopefully you don't have to take
such an extreme measure.
