rare cell population enumeration - CD34

I would be interested in anyone who is involved in accurate
enumeration of rare cell populations, especially CD34, in which the
frequency of occurrence is on the order of 1% or less and the
distribution of fl intensity is on the "low" end, making it
difficult and subjective to draw quad gates. In particular, any
novel approaches to population gating (or 'any' novel approaches
whatever) that remove some of the subjectivity would be of interest.
Pointers to methods in the literature that I have missed would be
helpful too.
