Dear colleagues,
When performing FACS analysis following BrdU labeling, can anything be
understood from the shape of the S phase profile? In control cells (HCT116 -
Human colon tumor cells), we see a tight uniform profile of cells entering and
exiting S phase - the classic U shape. In cells treated with compound, the
S-phase is more diffuse and has lost a tiny bit of fluorescence as compared to
control. Could this indicate variable rates of DNA synthesis or a block at the
entry to G2/M or does it mean nothing at all. Thanks in advance for any
responses.
