DNA content of Macrophage

Hello,
We are running samples for a student that are from lung lavages from
sheep. She is staining for DNA with PI and for antibodies with FITC. There
is a large population of macrophages in the lung samples. We have noticed
that the DNA content of the macrophage cells seems to be higher than that of
the lung cells. We were wondering if this is normal for macrophage cells, or
if it has to do with just those found in the lung? The student cultures the
lavage cells for about a week before they are stained for the antibodies and
fixed in 70% ETOH. She does try to deplete the macrophages from the culture,
but there are still a lot in there. In a separate culture, she grew only
macrophage cells, so they would not be able to ingest other cells, and we
still had the same results.
If anyone is familiar with looking at DNA content in macrophage
cells, we would appreciate any explanations!!
