The Course and Workshop will embrace basic and some selected
specialized techniques in:
- confocal imaging
- flow cytometry
- image analysis

There will be:

The Course is aimed at graduate students and young researchers from
areas of biology, biotechnology, medicine, chemistry, physics.

Participants are welcome to attend the lectures only (free of
charge) or register for practicals (tuition fee).

* LECTURES:

The lectures will cover:

CONFOCAL:
Principles of Confocal Microscopy
Choice of chromophores, laser lines, filters, lenses
in confocal microscopy
Photon statistics, sampling rate, confocal pinhole
- optimization of data collection
3-D image collection, restoration and analysis
Use of high NA oil and water immersion lenses
Resolution and contrast in confocal imaging

FLOW:
Principles of Flow Cytometry
Basic parameters: fluorescence, light scatter
Basic immunophenotyping using multicolour analysis
Cell cycle analysis, viability and apoptosis by flow
cytometry
Measurement of oxidative metabolism and cell
function
Flow measurements of platelet activation

IMAGE ANALYSIS:
Basic 2D image analysis
Image manipulation - noise, contrast, edge
enhancement, deconvolution

* PRACTICALS
------------
Practicals will cover:

CONFOCAL
Image acquisition - basic principles
Optimizing fluorescent image acquisition: role of
a confocal pinhole, sampling rate, etc.
Reflected light imaging
Multicolour image acquisition - bleedthrough
correction
Acquisition of 3D data sets

* Additionally:
We will offer a special tutorial on
microinjection of cells in culture

FLOW
DNA cell cycle analysis
Cell function by flow cytometry
Phenotyping
Multiparameter Data Analysis
Platelet activation

IMAGE ANALYSIS
Basic 2D image analysis
Image manipulation - noise, contrast, edge
enhancement
3-D image restoration and analysis
Deconvolution of confocal and wide field
fluorescence images

Participants of practicals/tutorials will gain hands-on experience
using:

>> a Bio-Rad MRC1024 confocal microscope
>> three Silicon Graphics workstations
>> BioRad, Beckton-Dickinson and Coulter flow
cytometers

The Course participants will study live and fixed
animal and plant cells.

Suggest your own topic for a tutorial

The prospective students are encouraged to suggest subjects of
practicals that they will find most useful (refer to the
Application Form).

Bring your own samples
=====================
The participants are welcome to bring their own samples to be
studied during practicals (no clinical material, please).

Please note:
Enrollment for practicals is limited to 20 students

* TUITION FEE and DEADLINES
---------------------------
Deadline for applications is March 15.

The tuition fee, payable to the Jagiellonian University, is:
$100,- for graduate students,
$150, - for faculty members
- if received before March 15, 1997

The tuition fees received after March 15 will be $150 and $200,
respectively.

The tuition fee includes the course materials, handouts, a ticket
for a banquet. Hotel, meals, etc. are not included.

* FINANCIAL SUPPORT, STIPENDS

Financial support will be available to participants who registered
for the Course. Please contact dr Dobrucki for details.

All students who successfully completed practicals on confocal
microscopy will be given 15 hours of use of the BioRad MRC1024
confocal microscope at the Jagiellonian University - free of
charge. These students will be welcome to continue their research
projects immediately following the Course or return at a later
date.

* ACCOMMODATION
---------------
A number of reasonably priced ($25-50//night) rooms have been
booked for the Faculty and Students at the Guest Houses of the
Jagiellonian University. Advance booking is required. Please
contact dr Dobrucki for details.

Please note:
June is a high tourist season in Krakow and only the pricey hotels
may have vacancies! Moreover, the Course coincides with a visit of
Pope John Paul II in Krakow. Early booking for University Guest
Houses is strongly recommended.

* COURSE CERTIFICATES
---------------------
Those who attended all Course lectures and successfully completed
at least four selected practicals\tutorials will be entitled to
receive a Course Certificate.

Museum of the Jagiellonian University (Collegium Maius)
featuring a unique collection of scientific equipment,
including old microscopes, copies of astronomical
instruments used by Nicolaus Copernicus, and more.
The medieval Salt Mine at Wieliczka
The Tatra Mountains
Krakow Old Town and the Royal Castle of Wawel

We are also planning a trip to a Museum at Auschwitz-Birkenau
(Oswiecim-Brzezinka) concentration camps.
