Dave Coder's response neglected one key aspect of Bjarne's original
question. It appears from the question that Bjarne is acquiring
*peak* fluorescence signals. However, Absolute fluorescence (e.g.
receptor number) is best measured on *integrated* fluorescence
signals, where the size of the cell (compared to the laser beam) is
not a factor. Newer FACScans and, I suspect, the FACSort have the
capacity to measure integrated fluorescence. With that caveat, I
agree with Dave's suggestions.
