Can anyone recommend protocols and reagents for enumeration of human CD34+
cells in cord blood, also in cytokine/factor stimulated cultured leucocytes
from
different sources. I am using a BD FACS Sort with a Mac (CellQuest
software),
and am a newcomer to this setup and FACS in general. In preliminary
experiments
I have used BD RPE-anti-CD34 and BD FITC-anti-CD45 - still playing around
with
settings - lots of questions, but getting fascinated. Is CD45 an optimal
marker
for total leucocyte detection - should I be going for a panel of
lineage-specific markers - any recommendations and advice on this would be
appreciated.
Also - I have years of experience with PC's (DOS, Windows) but am new to
Macs -
is there any way of saving files on Mac formatted discs then reading them in
PC's, and any PC software (especially downloadable shareware) for off-line
analysis of FACS data?? We may eventually have the Mac on a LAN with the
PC's,
but meantime what is the best way of getting the Mac data into a PC?
