We've been using the FITC-dUTP variant of the TUNEL assay (paraformaldehyde
fixed cells stored in PBS) to study granzyme/perforin mediated apoptosis.
We've recently noted an unexpectedly high percentage of cells being stained
in our negative control populations (yielding a bi-modal populations).
There are only two recent changes:
1) a new lot of 4% paraformaldehyde
2) repeated freeze-thaw cycles of the FITC-dUTP stock.
any help would be greatly appreciated
