Dear cytometrist?
I have been given this email address by Coulter Australia, in attempt to
find
a bulletin board that might assist with some technical aspects regarding
my
group getting started in some aspects of cell sorting. We have just
purchased
a Coulter Elite within the veterinary school and intended usage includes
some
clinical as well as basic research.

I have a question with regard to live sperm sorting. Does anyone have
experience with using the new Enterprise 50 mWatt UV laser for this
purpose?
The older literature cites using argon ion lasers running at about 200
mWatt
output power, but I am unaware whether the newer, smaller, (cheaper) UV
laser
is suitable for this purpose.

In response,
I have not used the Enterprise laser for sorting but understand from
talking to Dr Graeme Chapman that there may be some concerns regarding
its use for DNA. There are some UV capable lasers available for purchase,
from Uni NSW (one laser) and Uni Auckland (two lasers). The MAF at
Ruakura, NZ have used a FACS 440 for sorting Bull sperm, so it may be
worth talking to them. I don't have names for you but could possibly get
them if required.
