In our experience, using Supertechs FITC-conj. HT-6 monoclonal, Ortho
Permeafix does not allow for accurate TdT staining in the majority of
AMLs. It works very well in ALL but the typically dim fluorescence found
in AML cells presents a problem. We are routinely using the Caltag
method. I have done a lot of comparative studies with various fixation
methods and find Caltag to be the most reliable method (it also preserves
scatter characteristics very well) in comparison to the slide technique
and biochemical assays.
