I'm not sure that this is the right place to post this question
but here goes. We have used in situ hybs to localise Murray
Valley encephalitis virus RNA in the lymph nodes of mice
infected IP with MVE. Only a small population of cells
within the lymph node are positive for viral RNA and the
morphology suggests it may be the follicular dendritic
cells. I am interested in using flow cytometry to analyse
infected lymph node cell suspensions(or maybe just
peritoneal washouts?)to see if it is the dendritic cells
which are replicating virus or if other cell populations
are also involved in viral replication. Is this feasible?
and if so can anyone recommend some publications which
would be helpful. I'm sure it is quite obvious by now
that I have had no cytometry experience whatsoever. Any
advice would be appreciated.
