We used to preincubate MNC with 100 microliters of human Ig (3 mg/ml, outdated
vial of Sandoz IvIg) for 5-15 min, remove the supernatant, and then add the
Ab of interest. I found this much easier and safer to handle then human serum.
It worked wonders in blocking non specific binding, of even the stickiest
MoAb, such as the IgG2a class. Worth a try.
