help ? Apoptosis in flow vs Chromium release kill assay

Hi flowers ,

is there anybody out there doing apoptosis assay by flow to assess cytotoxic
effects on target cells ?

We normally do that by chromium release in a 3 hr assay, but a in special
case we find only few percent specific lysis on target cells even with a
high E:T ratio.

Increasing time of Chromium release assay leads to high spontaneous release
resulting in loss of adequate results.
In our case this poor specific lysis is no basis for further analysis of
specifity of the observed lysis.

What is your experience with apoptosis assay ( Annexin V ) in flow to be
compared with chromium release ?

What E:T ratios to start with ( more or less the same as in Chromium release ?)
What about incubation time ?
