According to the paper by Doornbos et al. (Cytometry 15, pp. 267-271
(1994) the TO-PRO-3 dye may be useful to resolve cell cycle
distributions by flow cytometry. The TO-PRO-3 dye is not membrane
permeant. If you are going to use this dye in conjunction with
antibodies you may have to tinker around with cell fixation /
permeabilization procedures. I am not sure that TO-PRO-3 is specific
for DNA: maybe you need to digest RNA in order to get a sharp CV.
Perhaps Harry Crissman (Los Alamos) has some useful experience with
this dye.
