Re: Hoechst Red Fluorescence

Ayala Sharp asks about red fluorescence from Hoechst 33342 which she saw
reported in a really interesting stem cell paper by Goodell et al. (J. Exp.
Med. Vol. 183 p. 1797). I have often wondered how many investigators have
been confounded by this not-so-commonly known dye property. If you look for
it, you'll see it before you can say "Hoechst."

There is some prior literature on the matter, and while the fluorescence is
easy to see, its' magnitude has some curious dynamics that are not well
characterized. It is the basis of my favorite apoptosis assay, and
interested readers might look at Chiu et al., (1996) J. Immun. Meth.
189:157-171.
