The new Cy7 tandems may provide the non-PerCP solution that people are asking
for. The first option is to use FITC, PE, Cy5PE, and Cy7APC--there should be
little compensation required between Cy5 and Cy7 (although there will be some).
Cy7APC was recently described in Cytometry articles by Andy Beavis and one from
our group (July and August of this year).

Another option is to use FITC, PE, and Cy7PE for the first laser, and APC, Cy5,
or Cy7APC for the second laser. The Cy7PE is very nice because it is a
reasonably bright first laser dye, and does not have substantial excitation by
the second laser. Cy7PE was described in our paper in Cytometry.

People wishing to use these dyes should send mail to the reagent manufacturers!
Both PharMingen and BD have successfully synthesized some of these dyes. These
tandems are not particularly difficult to make (detailed protocols for the
synthesis of these dyes are given in our Cytometry article), but they ARE very
useful. If you are interested, you can also send me EMail; if you have an
antibody that I am interested in, I may conjugate it for you (in return for
keeping half).
