Hi,
Here is a summary of the responses to my questions about APC conjugation.
If you want to conjugate antibodies to APC in your lab, APC and purified
APC are available through Molecular Probes and Cyanotech. One person wrote:
Molecular Probes (tel #(541) 465 8300) sells allophycocyanine
(cat #803) in native form or chemically stabilized by crosslinking
(cat #819). The crosslinked form does not dissociate at dilute
concentration and it is the form that we use to make our own
conjugates. We can prepare the pyridyldisulfide derivative of
either forms as custom synthesis and send a protocol on how to
perform the conjugation.
Apparently, the conjugation with APC can be tricky. For bright antigens,
a good alternative is using Cy5 (kits from Amersham), or one of the long
wavelength BODIPY dyes from Molecular Probes to make antibody conjugates.
There are some companies that will perform custom APC conjugations. They
are: 1. Chromaprobe in Mountain View, CA. The address for Malcolm Munn,
the owner, is listed at the web site
http://home.earthlink.net/~sukpark/vendors.html
2. BDIS has a custom conjugation program.
3. Pharmingen can also do custom conjugations.
Caltag, Pharmingen and BDIS alo offer APC conjugated antibodies.
Hope this is helpful,
