The Rainbow Calibration Particles from Spherotech (available
through Pharmingen, 1-800-848-6227) have worked well for us for
years. They have a dye mix in them which excites at UV, 488nm,
and dye laser emmission. Using standard Texas Red and APC
collection optics we regularly measusre cv's of less than 7%
with 598 nm dye laser excitation. The intensity in these
channels is approximately two log decades above the background
for unstained lymphoid cells. With 488nm excitation signal
intensities are similar to the dye laser measurements with
better cv's.
