I have a user that wants to couple protein-ligands to fluorescent latex
particles (Molecular Probes Fluospheres 0.01 um) and use them in binding
studies to the receptor on the cell-surface.
Is there anybody out there who has experience with this ?
Questions are in particular:
1. How to couple the protein to the fluospheres (Molecular Probes didn't
include enough information on their datasheets - Martin let me know if you
have more)...
2. How to block the free binding sites (BSA in excess ?)
3. What's the best size for the particles (is 0.01 um to small ?)
4. How to wash the cells or not before the acquisition ?
etc...
Anyway, any hints would be appreciated...
