To throw in my two cents worth:

I have been involved with flow cytometry since 1986 and during that whole
period have used, and have instructed the technicians I have taught to
use, 2.5% calf bovine serum in PBS for washing and final resuspension of
samples to be analyzed. Of course, we have gotten consistently good
results doing this, or we wouldn't have continued to do it. We normally
get tight population distributions, whether looked at as contour or dot
plots or as histograms, unless other factors such as instrumental or cell
sample quality problems are present. Our printouts are "publication
quality" and appear in many journal articles.
