1. Facilities: Labs are located in Michigan Cancer Foundation, a Wayne State University Medical School affiliate. Analytical Cytometry facility includes instrumentation labs, computer facility, tissue culture facility, space for visiting scientists.
2. Instrumentation:
Flow cytometry: Becton-Dickinson FACStar cell sorter with innova 90 argon laser; EPICS 753 dual laser cell sorter with 2 innova 90 argon lasers and CR559 dye (Rhodamine 6G) laser. Also available is a Becton-Dickinson FACScan 5-parameter flow cytometer.
Other major items: Olympus and Leitz phase/fluorescence microscopes equipped with CCD, Video, and standard camera systems. Perkin Elmer LS-3B fluorescence spectrometer, SLM-Aminco 8000C dual beam fluorescence spectrometer, Millipore Cytofluor 2300 multi-position plate reader, Zeineh Scanning Video Densitometry system.
Data acquisition and analysis: HP 9000 series systems, running Consort30, LYSYS II, and Paint-A-Gate Plus (automatic 3-color analysis with spreadsheet-compatible output); MDADS and EASY2 systems; several MS-DOS systems offering MODFIT and ISOCONTOUR (Verity), Elite (Coulter), MultiCycle (Phoenix), along with other graphics and statistics programs (CorelDraw, Picture Publisher, Sigma PLOT, InSTAT QuattroPRO, and Excel).
3. Specialization Areas: Extensive experience in measurement of functional parameters of drug resistance via dye/drug efflux assay in both solid and hematopoietic tissues. Modulation of efflux-related resistance with efflux blocking agents. Immunofluorescent determination of p-glycoprotein expression (C219, MRK16). Measurement of other mechanisms of resistance; DNA topoisomerase II, bc1-2, and MDR1 expression; glutathione levels. Immunofluorescence and DNA content analysis expertise.
