Concerning labeling of bacteria with FITC you first need to be more specific
about your "labeling" procedure. Are you labeling with FTIC conjugated Abs or
are you trying to directly conjugate the flurochrome to the bacterial surface
protein? You should seek advise from Howard Shapiro and get a copy of his
latest book since you state that you are new to cytometry. He has done
considerable work with bacterial cytometry and is probably the most
authoritative source on this subject as well as numerous other topics in
cytometry. He's not hard to locate and you will find him here quite often.
We have a list of most papers published on the subject of bacterial cytometry
dating back to light scattering observed by tyndall and will be glad to give
you a copy if you like. I think that you will have to determine the answers
to most of your own question with respect to bacterial cytometry since this
is a vastly unexplored area of cytometry. Hope this helps.
