Greetings,

If...
your laboratory performs CD34 enumeration on stem cell harvests as a
"billable" cytometry assay,

and...
you would be willing to send me a protocol of your strategy and
methodology,

I'd very much appreciate hearing from you.

Thanks in advance.

Hi Marc!
My laboratory has just started offering CD34 enumeration as a
"billable test" for the BM Transplantation service. We are not only
performing CD34 quantification on the harvests but also prior to the
harvest following GMCSF Tx to determine optimal time to begin
harvest. We will also monitor patients post transplantation. We have
set up the procedure using B-D's ProCount kit which is a three color
assay using a nucleic acid dye for determining nucleated cells (emits
in the FL-1;FITC range), CD45 PerCP, and CD34 PE. It also includes
a 3-color isotype control reagent (nucleic acid dye /IgG1-PE/
CD45-PerCP). The kit also contains a tube (True Count) of a known number of
lyophilized fluorescent beads which allows you to determine the
absolute number of CD34 cells/ul.
With regard to "billable", we have used the standard flow CPT code
88180 for each antibody marker. As long as your discharge code
(ICD-9) is a transplantation code you will be reimbursed.
I hope this information is helpful. If you have any other questions
feel free to address them specifically to me or call me at
(516)444-2373. Good Luck! Joanne Thomas
