Measurement of cellular DNA content and especially analysis of the cell division cycle have become among the most popular applications of flow cytometry. Potential uses include the determination of ploidy (often with significant implications for such areas as clinical tumour prognosis), analysis of cell division kinetics (e.g. S-phase fraction, doubling time, mitotic index, etc.), DNA Index measurement, flow karyotyping, and flow sorting of individual chromosomes, to mention but a few. However, such measurements often can be problematic and do require strict quality control. It is hoped that the material included in this section may serve to assist readers who might and who do work in this important field of flow cytometry.
